Radiobiological Studies of Vaccinia Virus.

چکیده
Radiation studies of vaccinia virus were carried out using two types of radiation, Coao gamma rays and ultraviolet light (UVL). When virus particles were irradiated with gamma rays a two-component survival curve was obtained. The capacity of Earle’s L cells to support the growth of vaccinia virus was relatively sensitive to gamma rays. Infectious centres formed by infecting L cells with vaccinia virus were irradiated at various times during the eclipse period of virus growth (the Luria-Latarjet experiment). When irradiated with gamma rays, infectious centres exhibited an initial sensitization to inactivation which persisted for at least 3 hours. At times later than 3 hours after infection the infectious centres became progressively more resistant to inactivation by gamma rays (“stabilization”) ; the survival curves remained almost single-hit throughout the eclipse period. Studies with FUdR, an inhibitor of DNA synthesis, showed little stabilization in the absence of viral DNA synthesis. Free particles of vaccinia virus were inactivated according to single-hit kinetics by UVL. The capacity of L cells to form virus was relatively resistant to UVL. In the Luria-Latarjet experiment using UVL, the survival curves of the infectious centres remained single-hit for the first 3 hours of the eclipse period, with slopes about half that of the survival curve for the free virus. Starting between 3 and 4 hours after infection, the survival curves became multihit, but their slopes remained parallel to the slopes of the survival curves of the early infectious centres. Possible interpretations of these results are discussed.

Vaccinia virus replication takes place in the cytoplasm of the host cell. The nearly 200 kbp genome owes part of its complexity to encoding most of the proteins involved in genome and mRNA synthesis. The multisubunit vaccinia virus RNA polymerase requires a separate set of virus-encoded proteins for the transcription of the early, intermediate and late classes of genes. Cell fractionation studi...

Foreign DNA was inserted into two nonessential regions of the vaccinia virus genome by homologous recombination in cells infected with virus and transfected with plasmids containing the foreign DNA elements flanked by vaccinia virus DNA. Thymidine kinase-negative (TK-) recombinants were selected after inserting foreign DNA into the coding region of the TK gene of wild-type vaccinia virus; TK+ r...

The interest in the action of chemical agents upon vaccinia virus is not of particularly recent origin. Jenner"6 suggested that zinc sulphate and copper sulphate could be used to mitigate or stop the progress of vaccinia infection. However, it was not until much later that Copeman"1 12 began a thorough study of the action of various chemical agents upon virus-containing calf lymph. This study w...

The development of a subunit vaccine for smallpox represents a potential strategy to avoid the safety concerns associated with replication-competent vaccinia virus. Preclinical studies to date with subunit smallpox vaccine candidates, however, have been limited by incomplete information regarding protective antigens and the requirement for multiple boost immunizations to afford protective immun...

Previous studies demonstrated that antibodies to live vaccinia virus infection are needed for optimal protection against orthopoxvirus infection. The present report is the first to compare the protective abilities of individual and combinations of specific polyclonal and monoclonal antibodies that target proteins of the intracellular (IMV) and extracellular (EV) forms of vaccinia virus. The ant...
